Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Analytical methods for lyophilized solids must account for the low moisture content and the fragile cake. Karl Fischer titration is widely used for water content, while near-infrared spectroscopy can measure moisture non-destructively in sealed containers. X-ray diffraction and modulated differential scanning calorimetry help identify crystalline or amorphous phases. Residual solvent analysis may be needed if organic solvents were used during formulation. The combination of these methods supports batch release and long-term stability assessment.
Lyophilized products are typically stored as sealed solids in vials or syringes. Moisture ingress is a major concern because many dried cakes are hygroscopic and can lose stability when exposed to humid air. Storage temperature depends on the formulation; some products are kept refrigerated, while others are stable at room temperature. Container closure integrity and headspace moisture are often monitored. Light protection may also be required for some photosensitive materials.
| Property | Value | Notes |
|---|---|---|
| Residual moisture | 0.5-3% w/w | Typical range for many biopharmaceuticals |
| Typical storage temperature | 2-8 °C | Some products require -20 °C or lower |
| Reconstitution time | Seconds to several minutes | Depends on cake porosity and diluent |
| Common moisture method | Karl Fischer titration | Measures water content in the solid |
| Container closure | Stoppered vial with seal | Protects against moisture and oxygen ingress |
Regulatory expectations for lyophilized products focus on consistent manufacture and documented stability. Batches are often monitored for moisture, appearance, potency, and sterility where applicable. Process parameters such as shelf temperature, chamber pressure, and drying time are recorded and controlled within validated ranges. Open questions remain about how best to predict long-term stability from short accelerated studies, especially for complex biologics. Variations in freezing rate and ice crystal size can produce differences that are not always visible but may affect performance.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture uptake. Residual water content is measured because small changes can alter chemical stability and cake appearance. Storage temperature depends on the material; many biological products are kept at 2–8 °C, while some require −20 °C or colder. Exposure to ambient humidity during handling can cause the porous solid to absorb water and collapse. Container closures and stoppers are therefore selected for low moisture transmission and compatibility.
Quality assessment of a lyophilized product includes cake appearance, residual moisture, reconstitution time, and container closure integrity. A uniform, porous cake suggests that freezing and drying stayed within the formulation's design space. Cracks, shrinkage, meltback, or a glassy film can indicate thermal abuse or a formulation problem. Analysts also test for subvisible particles and sterility when the product requires those specifications. Visual inspection alone cannot confirm biological activity or chemical stability, so it is combined with analytical methods.
Stability programs monitor lyophilized products under defined temperature and humidity conditions over time. Real-time studies at recommended storage conditions are the reference, while accelerated studies provide early signals of degradation pathways. Because a dry cake can still undergo oxidation, hydrolysis, or aggregation, stability depends on residual moisture, excipients, and container headspace. Open questions include how best to predict long-term stability from short accelerated runs and how vial-to-vial variability affects shelf life. Current guidance treats these predictions as product-specific rather than universally generalizable.
Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.
Quality control also examines cake structure, color, and reconstitution behavior. A collapsed or shrunken cake can indicate a thermal excursion during drying. Analytical methods such as X-ray diffraction, differential scanning calorimetry, and near-infrared spectroscopy can detect crystallinity or moisture distribution. Regulatory expectations focus on validated assays and lot-to-lot consistency. Questions remain about how well accelerated stability tests predict long-term behavior for every formulation. Visual inspection remains common but is subjective without trained reviewers and reference images.
After lyophilization, a product's quality depends on residual moisture, cake appearance, and reconstitution time. Residual moisture is often measured by Karl Fischer titration or thermogravimetric analysis. A low moisture content can slow chemical degradation, but overly dry cakes may be brittle or slow to dissolve. Stability studies track these attributes over months under defined temperature and humidity conditions. Batch records link these measurements to specific process runs and help identify trends before a product fails specification.
With alprazolam, a short-acting benzodiazepine, taken for eight weeks, 65% of patients experienced significant rebound anxiety. To some degree, these older benzodiazepines are self-tapering. The benzodiazepines diazepam and oxazepam have been found to produce fewer withdrawal reactions than alprazolam, temazepam, or lorazepam. Factors that determine the risk of psychological dependence or physical dependence and the severity of the benzodiazepine withdrawal symptoms during dose reduction of alprazolam include: dosage used, length of use, frequency of dosing, personality characteristics of the individual, previous use of cross-dependent/cross-tolerant drugs (alcohol or other sedative-hypnotic drugs), current use of cross-dependent/-tolerant drugs, use of other short-acting, high-potency benzodiazepines, and method of discontinuation.
=== Provisions on agriculture === The law rescinds unobligated IRA funding for the Conservation Stewardship Program, Environmental Quality Incentives Program, Agricultural Conservation Easements Program, and Regional Conservation Partnership Program, and adds it to the USDA budget baseline. The OBBBA outlaws climate action-related goals for agriculture. The law also lifts income caps on households that rely on agriculture for more than 75% of their income, potentially empowering corporate farming. The law raises reference prices under the Price Loss Coverage and Agricultural Risk Coverage programs, resulting in $54 billion in additional spending over 10 years. The law increases spending on crop insurance programs by $6.3 billion over 10 years and disaster relief programs at USDA by $2.9 billion in the same timeframe.
TikTok has since apologized and instituted a ban against anti-LGBTQ ideology, but censorship continues on Douyin due to regulations in China. Douyin guidelines also forbid live broadcasting by unregistered foreigners, "feudal superstition", "money worship", smoking and drinking, competitive eating by the "already obese", "toxic" slime, "pornographic" ASMR such as ear-licking, and female anchors wearing revealing clothes. ByteDance said its early guidelines were global and aimed at reducing online harassment and divisiveness when its platforms were still growing. They have been replaced by versions customized by local teams for users in different regions. A March 2021 study by the Citizen Lab found that TikTok did not censor searches politically but was inconclusive about whether posts are. A 2023 paper by the Internet Governance Project at Georgia Institute of Technology concluded that TikTok is "not exporting censorship, either directly by blocking material, or indirectly via its recommendation algorithm." After increased scrutiny, TikTok said it is granting some outside experts access to the platform's anonymized data sets and protocols, including filters, keywords, criteria for heating, and source code.
The Pd2(dba)3/MePhos catalytic system has been applied to late stage Suzuki cross couplings. This reaction has been conducted on a kilogram (2.2 lbs)scale, and no specific palladium-removal treatment was required as the excess imidazole present in the final amide coupling step coordinated to the Pd and generated a removable byproduct.
=== Other === Overactive TGF-β pathway, with an increase of TGF-β2, was reported in the studies of patients with keratoconus. There is substantial evidence in animal and some human studies that TGF-β in breast milk may be a key immunoregulatory factor in the development of infant immune response, moderating the risk of atopic disease or autoimmunity. Skin aging is caused in part by TGF-β, which reduces the subcutaneous fat that gives skin a pleasant appearance and texture. TGF-β does this by blocking the conversion of dermal fibroblasts into fat cells; with fewer fat cells underneath to provide support, the skin becomes saggy and wrinkled. Subcutaneous fat also produces cathelicidin, which is a peptide that fights bacterial infections.

Mahathir's political views have shifted during his lengthy career. During the 1980s and 1990s, he was a supporter of third-worldism, while during other periods he has been a proponent of "Asian values" and globalisation. A Muslim thinker, he holds Islamic political views. In 2002, he characterised himself as an Islamic fundamentalist. Mahathir is generally respected in developing and Islamic countries, particularly due to his oversight of Malaysia's economic growth and his support of liberal Muslim values. Mahathir has been described as a proponent of Malay nationalism. In The Malay Dilemma, he argued that the Malay race had been marginalised, and voiced his support for affirmative action policies for them. Upon his first resignation, he expressed his disappointment at the progress made towards his "principle task" of supporting the Malay race. In 2021, Mahathir said he did not believe in Ketuanan Melayu, calling it a "fantasy", and said instead that he believed in the concept of "Bangsa Malaysia". He has been described as anti-royalist by the left-wing French newspaper Libération, owing to his efforts to oppose immunity for members of Malaysia's monarchies. Mahathir is a vocal critic of neoliberalism and the Western world. In 2011, Mahathir suggested that the September 11 attacks might have been staged by the United States government. Mahathir condemned the Universal Declaration of Human Rights in 1997, suggesting it be revised to place greater importance on economic growth over civil liberties.
=== Carcinogenicity === The National Toxicology Program from the Department of Health and Human Services published the Fifteenth Edition Report on Carcinogens, which stated that BaP caused tumors in 8 species, including nonhuman primates.
=== "...eine neue Reaktion des Kreatinins" === Creatinine was first synthesized in vitro by Ivan Horbaczewski in 1885. One year later, Jaffe's research was published in the paper Über den Niederschlag, welchen Pikrinsäre in normalem Harn erzeugt und über eine neue Reaction des Kreatinins. Jaffe had noticed that, when mixed in a sodium hydroxide (NaOH) solution, picric acid and creatinine formed a reddish-orange color and needle-like crystal precipitate. By using zinc chloride in a process known as the Neubauer reaction, and then performing the Weyl's test, a colorimetric reaction using sodium nitroprusside (SNP), he determined that the precipitated compound was a double salt of the solution. Although he found the amount of precipitate directly proportional to the creatinine concentration, he also noted that the reaction was highly nonspecific and could be observed with many other organic compounds.

Mescaline is biosynthesized from tyrosine, which, in turn, is derived from phenylalanine by the enzyme phenylalanine hydroxylase. In Lophophora williamsii (peyote), dopamine converts into mescaline in a biosynthetic pathway involving m-O-methylation and aromatic hydroxylation. Tyrosine and phenylalanine serve as metabolic precursors towards the synthesis of mescaline. Tyrosine can either undergo a decarboxylation via tyrosine decarboxylase to generate tyramine and subsequently undergo an oxidation at carbon 3 by a monophenol hydroxylase or first be hydroxylated by tyrosine hydroxylase to form L-DOPA and decarboxylated by DOPA decarboxylase. These create dopamine, which then experiences methylation by a catechol-O-methyltransferase (COMT) by an S-adenosyl methionine (SAM)-dependent mechanism. The resulting intermediate is then oxidized again by a hydroxylase enzyme, likely monophenol hydroxylase again, at carbon 5, and methylated by COMT. The product, methylated at the two meta positions with respect to the alkyl substituent, experiences a final methylation at the 4 carbon by a guaiacol-O-methyltransferase, which also operates by a SAM-dependent mechanism. This final methylation step results in the production of mescaline. Phenylalanine serves as a precursor by first being converted to L-tyrosine by L-amino acid hydroxylase. Once converted, it follows the same pathway as described above.
The first metallocene was ferrocene, discovered simultaneously in 1951 by Kealy and Pauson, and Miller et al. Kealy and Pauson were attempting to synthesize fulvalene through the oxidation of a cyclopentadienyl salt with anhydrous FeCl3 but obtained instead the substance C10H10Fe At the same time, Miller et al reported the same iron product from a reaction of cyclopentadiene with iron in the presence of aluminum, potassium, or molybdenum oxides. The structure of "C10H10Fe" was determined by Geoffrey Wilkinson et al. and by Ernst Otto Fischer et al. These two were awarded the Nobel Prize in Chemistry in 1973 for their work on sandwich compounds, including the structural determination of ferrocene. They determined that the carbon atoms of the cyclopentadienyl (Cp) ligand contributed equally to the bonding and that bonding occurred due to the metal d-orbitals and the π-electrons in the p-orbitals of the Cp ligands. This complex is now known as ferrocene, and the group of transition metal dicyclopentadienyl compounds is known as metallocenes. Metallocenes have the general formula [(η5-C5H5)2M]. Fischer et al. first prepared the ferrocene derivatives involving Co and Ni. Often derived from substituted derivatives of cyclopentadienide, metallocenes of many elements have been prepared. One of the very earliest commercial manufacturers of metallocenes was Arapahoe Chemicals in Boulder, Colorado
== Target and events == The primary target of the doctrine was originally the largest communist country, the Soviet Union. However, additional targets also included the other communist countries like the China, Laos, Vietnam, and North Korea. After the dissolution of the Soviet Union in 1991 and China's economic prominence in the early 21st century, China became the major target of the doctrine.
==== Early 1990s ==== The Virgilio Barco Vargas (1986–1990) administration, in addition to continuing to handle the difficulties of the complex negotiations with the guerrillas, also inherited a particularly chaotic confrontation against the drug lords, who were engaged in a campaign of terrorism and murder in response to government moves in favor of their extradition overseas. In June 1987, the ceasefire between FARC and the Colombian government formally collapsed after the guerrillas attacked a military unit in the jungles of Caquetá. According to journalist Steven Dudley, FARC founder Jacobo Arenas considered the incident to be a "natural" part of the truce and reiterated the group's intention to continue the dialogue, but President Barco sent an ultimatum to the guerrillas and demanded that they immediately disarm or face military retaliation. Regional guerrilla and Army skirmishes created a situation where each violation of the ceasefire rendered it null in each location, until it became practically nonexistent. By 1990, at least 2,500 members of the FARC-founded Patriotic Union had been murdered, according to historian Daniel Pecáut, which led up to that year's assassination of presidential candidate Bernardo Jaramillo Ossa.

Residual moisture can influence chemical degradation, cake collapse, and long-term stability. Low moisture levels usually improve stability, but each product has an optimal range.
Most lyophilized products are stored upright at controlled temperatures, often refrigerated or frozen. Protection from moisture and light helps maintain the dried cake.
A diluent is added to the dried cake, which dissolves to form a solution or suspension. Gentle mixing avoids foaming and preserves sensitive molecules.
Most lyophilized products are stored in sealed containers at controlled temperature and humidity. Some require refrigeration, while others are stable at room temperature. Protection from light and moisture is often necessary.
