== Function == The UBC9 protein encoded by the UBE2I gene constitutes a core machinery in the cell's sumoylation pathway. Sumoylation is a process in which a Small Ubiquitin-like MOdifier (SUMO) is covalently attached to other proteins in order to modify their behaviour. For example, sumoylation may affect a protein's localization in the cell, its ability to interact with other proteins or DNA. UBC9 performs the third step in the sumoylation life cycle: the conjugation step. When SUMO protein precursors are first expressed, they first undergo a maturation step in which the four C-terminal amino acids are removed, revealing a di-glycine motif. In a second step, an E1 activating complex binds to SUMO at its di-glycine and passes it on to the E2 protein Ubc9, where it forms a thioester bond with a cysteine residue within Ubc9's catalytic pocket. The loaded Ubc9 is now ready to perform the sumoylation of its various target proteins (also called substrates). It recognizes a particular motif of amino acid residues in these substrates: A large hydrophobic residue, followed by a lysine, followed by a spacer, followed by an acidic residue. This motif is usually described in shorthand as ΨKxD/E. The central lysine within the substrate's recognition motif is inserted into the catalytic pocket. There the carboxyl terminus of SUMO's di-glycine forms a peptide bond with the ε-amino group of the lysine. This process can be assisted by an E3 ligase protein. === Initial 90-day freeze === On January 24, 2025, President Donald Trump ordered a near-total freeze on all foreign aid. In February, the administration placed most employees on administrative leave. The absence of authorization from Congress led to lawsuits against the Trump administration. Also in February, the administration made several allegations of wasteful spending and fraud, allegations which were generally reported to be false. Several days later, Secretary of State Marco Rubio issued a waiver for humanitarian aid. However, a key issue developed over whether the waivers for lifesaving aid were actually translating into aid flowing. Despite the waiver, there was still much confusion about what agencies should do. More than 1,000 USAID employees and contractors were fired or furloughed following the near-total freeze on U.S. global assistance that the second Trump administration implemented. On January 27, 2025, the agency's official government website was shut down.
=== Pharmacokinetics === Piperacillin is generally available in their stable form as crystallized potassium or sodium salt, quickly losing bactericidal activity upon dissolution due to their short half-lives. As the gastrointestinal tract does not absorb piperacillin and tazobactam, they are dissolved in a solution before being administered to a patient, through parenteral means. Excreted through renal mechanisms like glomerular or tubular filtration as a component of urine, uncontrolled dosages of the drug can cause renal dysfunction and competitive inhibition of excretion, delaying piperacillin-tazobactam excretion, and endangering patients to drug exposure.
The making and drinking of cider is traditional in several areas of northern Spain, mainly Galicia, the Principality of Asturias, Cantabria, and the Basque Country. The largest producer of cider in Spain is the Atlantic region of Asturias, where cider is considered not only a beverage but an intrinsic part of its culture and folklore. Asturias amounts more than 80% of the whole production of Spain. The consumption of cider in Asturias is of 54 litres per person/year, probably the highest in any European region. The first testimony about cider in Asturias was made by Greek geographer Strabo in 60 BC. The traditional Asturian sidra is a still cider of 4–8% strength, although there are other varieties. Traditionally, it is served in sidrerías and chigres, pubs specialising in cider where it is also possible to have other drinks as well as traditional food. One of the most outstanding characteristics is that it is poured in very small quantities from a height into a wide glass, with the arm holding the bottle extended upwards and the one holding the glass extended downward. This technique is called escanciar un culín (also echar un culín) and is done to get air bubbles into the drink (espalmar), thus giving it a sparkling taste like Champagne that lasts a very short time. Cider is also poured from barrels in the traditional Espichas. One of the most popular ciders in Spain is called "El Gaitero" (the bagpipe player) which can be found everywhere in Spain and which is produced in this region.
=== Immunoprecipitation === The liquid-phase ligand binding assay of immunoprecipitation (IP) is a method that is used to purify or enrich a specific protein, or a group of proteins, using an antibody from a complex mixture. The extract of disrupted tissue or cells is mixed with an antibody against the antigen of interest, which produces the antigen-antibody complex. When antigen concentration is low, the antigen-antibody complex precipitation can take hours or even days and becomes hard to isolate the small amount of precipitate formed. The enzyme-linked immunosorbent assay (ELISA) or Western blotting are two different ways that the purified antigen (or multiple antigens) can be obtained and analyzed. This method involves purifying an antigen through the aid of an attached antibody on a solid (beaded) support, such as agarose resin. The immobilized protein complex can be accomplished either in a single step or successively. IP can also be used in conjunction with biosynthetic radioisotope labeling. Using this technique combination, one can determine if a specific antigen is synthesized by a tissue or by a cell.

Chief Royal Navy vessels deployed to the Persian Gulf included Broadsword-class frigates, and Sheffield-class destroyers. Other R.N. and RFA ships were also deployed. The light aircraft carrier HMS Ark Royal was deployed to the Mediterranean Sea. Several SAS squadrons were deployed. A British Challenger 1 achieved the longest range confirmed tank kill of the war, destroying an Iraqi tank with an armour-piercing fin-stabilized discarding-sabot (APFSDS) round fired over 4,700 metres (2.9 mi)—the longest tank-on-tank kill shot ever recorded.

=== Social programs === In July 2021, Moore said he opposed Governor Larry Hogan's decision to end expanded federal unemployment benefits provided by the American Rescue Plan Act of 2021 early. In December 2022, Moore said he supported indexing the state's maximum unemployment insurance payment to inflation. In August 2022, Moore supported protests led by veterans at the United States Capitol to pass the Honoring our PACT Act of 2022, which would provide benefits for veterans exposed to burn pits and other toxic phenomena. During his gubernatorial campaign, Moore said he supported establishing a state "baby bonds" program, which would cost roughly $100 million per year and be seeded with $3,200 for every child born on Medicaid, to target the racial wealth gap. If enacted, it would be the largest baby bond program ever enacted in the United States. In September 2022, he told the Maryland Family Network that he would support child care programs by subsidizing the service through tax credits for low-income families. Moore has not advocated for establishing a statewide baby bonds program since becoming governor, but has supported other measures toward expanding childcare access in the state such as an expansion of Maryland's child tax credit. During the 2024 legislative session, Moore introduced the ENOUGH Act, a bill to provide $15 million in grants toward underserved communities. The bill passed and was signed into law in May 2024.

=== Criminalization and medical use === Non-clinical use of opium was criminalized in the United States by the Harrison Narcotics Tax Act of 1914, and by many other laws. The use of opioids was stigmatized, and it was seen as a dangerous substance, to be prescribed only as a last resort for dying patients. The Controlled Substances Act of 1970 eventually relaxed the harshness of the Harrison Act. In the United Kingdom the 1926 report of the Departmental Committee on Morphine and Heroin Addiction under the Chairmanship of the President of the Royal College of Physicians reasserted medical control and established the "British system" of control—which lasted until the 1960s. In the 1980s the World Health Organization published guidelines for prescribing drugs, including opioids, for different levels of pain. With little or no scientific evidence to support their claims, industry scientists and advocates suggested that people with chronic pain would be resistant to addiction. The release of OxyContin in 1996 was accompanied by an aggressive marketing campaign promoting the use of opioids for pain relief. Increasing prescription of opioids fueled a growing black market for heroin. Between 2000 and 2014 there was an "alarming increase in heroin use across the country and an epidemic of drug overdose deaths". As a result, health care organizations and public health groups, such as Physicians for Responsible Opioid Prescribing, have called for decreases in the prescription of opioids.
